Isolated regulatory T cells (Tregs) are expanded in vitro using anti-CD3/anti-CD28 beads (activation/proliferation) and IL-2 (survival). Polyclonal Tregs are reintroduced into patients safely in graft-versus-host disease (GvHD), transplantation rejection, and type 1 diabetes mellitus (T1DM) clinical trials. The expansion step provides a window of opportunity for in vitro Treg manipulation to boost their function and stability, which are impaired in autoimmunity.
